countdown.wav is located at /Applications/RVGL.app/Contents/Resources/wavs on macOS.
I don't like the sound so I ran it through a low-pass filter in Audacity and replaced the original with my edit. I played it to ensure the file actually sounds different.
When I run the game, the sound is actually unchanged. What am I doing wrong?
Edit: I'm not using the game launcher. I just downloaded the game itself from the website.
Update: I got it to work using the launcher version of RVGL. I edited the wav at /Users/<me>/Library/Application Support/rvmm/packs/game_files/wavs/
[SOLVED] I edited countdown.wav, but in the game it sounds unchanged. What am I doing wrong?

Should something similar happen to someone else, I'd suggest to do a file search on your computer system. In the case of the start countdown, you are looking for "countdown.wav", which you would then find in multiple locations. Don't forget to search for hidden folders as well.
